ABSTRACT

The aim of this study was to establish a novel technology using microalgae for NO₃⁻ removal from high concentration wastewater and conversion to algal proteins. The effects of cultivation modes and illumination modes on the biomass yield, NO₃⁻ assimilation rate and algal protein yield were first investigated in shaking flasks for mixotrophic cultivation of Chlorella pyrenoidosa, and subsequently the scale-up verification in 5-L photo fermenter was successfully conducted. Fed-batch cultivation without medium recycling was the best cultivation mode in shaking flask system, in which the highest biomass yield (35.95 g/L), the average NO₃⁻ assimilation rate (2.06 g/(L·d)) and algal protein content (up to 42.44% of dry weight) were achieved. By using a staged increase of light intensity as illumination modes, the specific growth rate of cells could be significantly promoted to the highest (0.65 d⁻¹). After a 128-hour continuous cultivation in a 5-L photo fermenter, the highest biomass yield and the average NO₃⁻ assimilation rate were reached to 66.22 g/L and 4.38 g/(L·d) respectively, with the highest algal protein content at 47.13% of dry weight. Our study could provide a photo fermentation technology of microalgae for highly efficient treatment of waste industrial nitric acid and/or high concentration nitrate wastewater. This microalgae-based bioconversion process could coproduce protein-rich microalgal biomass, which facilitates the resource utilization of these type wastewater by trash-to-treasure conversion.

ABSTRACT

Colorectal cancer (CRC) is a common malignant tumor in the digestive tract, and 30%-85% of CRCs express epidermal growth factor receptors (EGFRs). Recently, treatments using cetuximab, also named C225, an anti-EGFR monoclonal antibody, for CRC have been demonstrated to cause an S492R mutation in EGFR. However, little is known about the biological function of S492R EGFR. Therefore, we attempted to elucidate its biological function in CRC cells and explore new treatment strategies for this mutant form. Our study indicated that EGFR and S492R EGFR accelerate the growth of CRC cells in vitro and in vivo and monoclonal antibody CH12, which specifically recognizes an EGFR tumor-specific epitope, can bind efficiently to S492R EGFR. Furthermore, mAb CH12 showed significantly stronger growth suppression activities and induced a more potent antibody-dependent cellular cytotoxicity effect on CRC cells bearing S492R EGFR than mAb C225. mAb CH12 obviously suppressed the growth of CRC xenografts with S492R EGFR mutations in vivo. Thus, mAb CH12 may be a promising therapeutic agent in treating patients with CRC bearing an S492R EGFR mutation.

ABSTRACT

Objective@#To investigate the development of hepatocyte senescence during liver fibrogenesis and to explore the effect and possible mechanism of insulin-like growth factor 1 (IGF-1) on hepatocyte senescence and liver fibrosis.@*Methods@#A total of 42 male Sprague Dawley (SD) rats were selected. Eighteen rats were induced by carbon tetrachloride (CCl4) to establish the rat model of liver fibrosis. On the day 0, six and 28 after the establishment of the model, six rats were executed respectively to analyze the liver fibrosis and hepatocyte senescence in CCl4-induced liver fibrosis rat models. Twenty-four rats were divided into control group, CCl4 group, CCl4+ lentivirus vector (LV-CTR) group and CCl4+ LV-IGF-1 group, with six rats in each group.The rats were sacrificed on the 28th day after the establishment of the model. The liver tissues were obtained and the inferior vena cava blood was collected to analyze the effect of IGF-1 overexpression on liver fibrosis and hepatocyte senescence. Analysis variance (ANOVA), least significant difference (LSD) and Dunnett T3 test were performed for statistical analysis.@*Results@#Steatohepatitis on the 6th day and early stage of hepatic fibrosis on the 28th day, which indicated the model was successfully established. The results of the effects of IGF-1 overexpression on hepatic fibrosis and hepatocyte senescence showed that on the 28th day, compared with those of control group, both the score of Ishak liver inflammation and necrosis and the score of Ishak liver fibrosis were increased in the CCl4 group, CCl4+ LV-CTR group and CCl4+ LV-IGF-1 group (0, 14.55±1.94, 15.43±2.19 and 10.29±1.47, respectively; 0, 3.51±0.51, 3.21±0.79 and 1.32±0.40, respectively). The area of liver tissues by Masson staining (0.45±0.40, 5.62±1.08, 6.03±0.65 and 2.88±1.54), SA-β-Gal staining (1.75±0.80, 4.28±1.19, 4.92±1.14, 3.11±0.79), p53 (2.02±0.81, 4.36±1.02, 4.72±0.72 and 3.58±0.70) and progerin (0.72±0.40, 4.52±1.01, 4.01±1.25 and 2.66±0.80) all were increased. The levels of serum IGF-1 all were decreased ((632.00±6.04), (503.00±40.42), (508.00±21.94) and (572.40±5.94) ng/L). However the levels of ALT all were increased ((11.20±5.97), (214.00±73.90), (245.00±76.06) and (30.00±5.00) U/L). The relative expression levels of p53 (0.58±0.06, 1.78±0.18, 1.72±0.10 and 1.23±0.22) and progerin (0.12±0.02, 0.78±0.15, 1.32±0.20 and 0.81±0.16) in the primary hepatocytes were increased. The differences were all statistically significant (F=91.674, 90.778, 32.982, 9.726, 10.640, 17.029, 103.910, 30.059, 64.707 and 97.457, all P<0.05). Compared with those of CCl4+ LV-CTR group, the score of Ishak liver inflammation and necrosis and the score of Ishak liver fibrosis were decreased in the rats′ liver tissues of CCl4+ LV-IGF-1 group, the areas of Masson staining, SA-β-Gal staining, p53 and progerin in the liver tissues were decreased, the level of serum IGF-1 was increased, the level of ALT was decreased, and the relative expression levels of p53 and progerin in primary hepatocytes both were decreased. The differences were all statistically significant (all P<0.05, respectively).@*Conclusions@#Hepatocyte senescence increases in the process of liver fibrosis induced by CCl4. Overexpression of IGF-1 may alleviate liver injury, improve hepatocyte senescence and liver fibrogenesis by regulating the nuclear p53/progerin pathway.

ABSTRACT

Objective: During the five years period from 2006 to 2010, the Chinese government had increased the special funding for primary maternal and child health enterprise to analyze the total amount and constitution of the special funding respectively. Methods: From the perspective of special funding investment in the eastern, central and western regions with different levels of economic development, to perform the comparative analysis. Results and Conclusion:The government’s overall investment on maternal and child special funds is in increasing trend, the average per capita amount of special funds invested in west region is higher than the eastern and central region; in the aspect of investment constitution , the central government special funds for women and children is the main source, which accounts for more than 85% of total investment, the reducing and eliminating items in eastern and western regions and the inpatient childbirth subsidy items constitute the major component of special funding for primary maternal and child health in corresponding region. Based on the analysis of investment on the condition of primary maternal and child health , policy recommendations for further development of the cause of women and children are provided.

ABSTRACT

Background and purpose: Primary study showed that clusterin was associated with tumorigenicity. The goal of the study is to investigate the role of secretory clusterin in non-small cell lung cancer cell A549/H460. Methods: The lung cancer cell A549/H460 was treated with purified secretory clusterin, the Boyden Chamber migration assay was used to detected the migration of the lung cancer cell;the CCK8 assay was used to detected the growth of the cells;microRNA expression spectrum in H460 treated with secretory clusterin was analyzed, after that, we used the real-time florescence quantification detected the expression of microRNA in H460, and the biological function of microRNA molecular mechanisms of secretory clusterin was analyzed. Results: Secretory clusterin promoted the migration in A549/H460 (P<0.000 1);Secretory clusterin inhibited the growth in H460/A549 (P<0.000 1);MicroRNA-302b-3p, microRNA-23a-5p and microRNA-101-5p was overexpressed in H460 when treated with secretory clusterin. Conclusion:Secretory clusterin could promote the migration and inhibit the growth of lung cancer cell;It could change the microRNA expression spectrum as well. Our studies revealed that secretory clusterin could be used as a tool for further study, and it is a potential target in the treatment of lung cancer.

ABSTRACT

Objective To investigate the effect of the UU and CT in the infertility.Methods Tractus genitalis secretion of 305 infecund women and 130 normal women were tested by realtime PCR and the UU and CT were analyzed.Results The infection rat of UU,CT and UU + CT in the test group were 59.3% 、41.0% and 17.0% which were all higher than that of control group(χ2 = 89.82、44.86、19.11,all P<0.05).However,the difference couldn't be seen between the primary infertility group and the secondary infertility group.Conclusion The women's infertility was related to the infection of the UU and CT.

Results: There were significant differences among Herba Leonuri Preparations from different pharmaceutical factories, place of origin and batches. Conclusions: It is suggested that the determination item of stachydrine hydrochloride be added.

ABSTRACT

AIM: To evaluate the long-term clinical efficacy of supplementary Danshen Tablet therapy in ischemic cardiomyopathy patients with heart failure. METHODS: 100 ischemic cardiomyopathy patients with heart fai-lure were randomly assigned into two groups,Danshen group(50 patients) and conventional therapy group(50 patients).The patients in Danshen group were administrated Danshen Tablet(three tablets a time,three times a day) plus conventional therapy for one year.NYHA functional class,6 min walking-distance,echocardiographic indices,major-adverse-cardiovascular-events(MACE) and mortality were evaluated before and after 1 year therapy.(RESULTS):(After) 1 year therapy,the improvements of NYHA functional class and 6 min walking-distance were more significantly effective in Danshen group than those in conventional group(P
